Playing as Carthage I came across 2 water supply building chains - desert water supply which only has 2 tiers and water supply which has 4, however for some towns I'm only able to build desert water supply (in africa) while others I can do both (in tripolitana), both have the same city building. wondering what's preventing me from building the water supply chain? Thanks much.

Pretty sure it's region dependent.

Some regions, particularly desert ones like Africa, only have access to desert water supplies. These give less benefits because less water in the desert. Quite a good way (imo) to reflect the differing building potentials of the different regions - if you want to build a big city in the desert then you will have to build more wells or deal with the higher risk of plagues.
