    For the first question:
    go into your medieval II folder, then data/world/maps/campaing/imperial campaing and find a file called descr_strat.txt (make a backup, youll thank me once for this), open it and find the city you want to change and then simply change the population to whatewer you like, see spoiler
    Spoiler Alert, click show to read:
    settlement castle
    {
    level large_town
    region Ragusa_Province

    year_founded 0
    population 1500
    plan_set default_set
    faction_creator venice
    building
    {
    type core_castle_building castle
    }
    building
    {
    type equestrian stables
    }
    building
    {
    type castle_barracks mustering_hall
    }
    building
    {
    type hinterland_castle_roads c_roads
    }
    building
    {
    type castle_port c_port
    }
    }

    for the second question:
    as far as i know, in vanilla you cannot change this, you can change this though in mods for M2TW
    Peace and prosperity to you noble sadiq
