Is there anyway to assasinate a prest? I remember being able to assisinate just about anybody but It wont let me use the assason to kill a prest. Can you do it with a gentleman?

Yes, use a rake (he wears a mask).

Right click on your target. A window should pop up mid screen about the target and percentage chance of killing. Click on little dagger button. Voila! Dead priest.

Even at 25% chance my rakes can 99% kill a priest, for other characters though, not so.

Rakes dont work, I tryed that allready, it wont let me even try and assasinate him, rake works on anyone else bu them.

Sometimes the game glitches and makes it impossible to assassinate a target. The same happens with my navies being unable to blockade a port, which gets to be very annoying, but it has not started happening until the latest patch was released. The cursor does not become a big X, it stays as the movement cursor, but I am unable to move there. Is that what your problem is like?

A. Make sure it is an enemy priest.

B. If its the problem thedukederek is describing (happens to me too) move a short distance away and click the target (usually works then) for assassination or blockade. Otherwise wait a turn and try again.
