I usually keep 2 to hold up my flanks and encircle my enemy and chuck grenades at them.
4 in my stacks to throw grenades over the heads of my line infantry
usually two units per stack, I keep them behind the main line until the other army moves in then it's grenade time. I never have them set at fire at will. i wait for the right moment to unleash them. Then i change back to rifle mode to add to the chaos.
I get about 2-4 if I can to lob nades and clear buildings.
Last time I tried to clear buildings with them nothing happened.
2 to 4 in a stack used as shock troops. Similar to how the No-Dachi Samurai were used in Shogun. Commit your forces to a goal, when it seems like there is a stalemate at some portion in your line, throw in the grenadiers to tip the balance.
I keep grenadiers aside for assault purposes only.
They are not part of the line, but may be used as part of a flank action.
I generally do not have them in my armies unless I have an urban target in mind, but I do value their service and am always impressed with how well they perform when brought into a fight as a closer.
Have you seen what 4 grenadiers approaching from a flank can do to a full unit of veteran line infantry with one grenade toss? Two grenade tosses and there are usually 3-4 survivors. So much so that I call the 4 grenadier platoons the "Flying Murder Squad".
I use my grenadiers mainly for their melee capabilities. After weakening an enemy unit, I usually bayonet charge, since grenadiers can usually defeat most forces even if they're outnumbered. Most important use for my grenadiers though, is to clear those pesky defenders that are garrisoned in houses if I don't have artillery.

I hold the Grenadiers in reserve for when the fighting gets really hairy, usually I'll throw grenades at their troops, switch to musketfire and shoot them in the back as they flee, then charge them.
Grenadiers have a lot of uses and most of them will include grnade throwing.
They good for flanking engaged enemy infantry line.
They good for throwing grenades from behind your infantry line, either between walleys, or you can just move your inf line almost at point blank to enemy line and have grenadiers start tossing grenades safely from behind line infantry backs.
They good for throwing grenades to fort wall defenders in seiges.
They good to throw grenades from fort wall at attakers in seiges.
They good for plugging holes in your line.
They good for bayonet charge in decicive moment of battle
They good for keeping them as reserve
They good for urban combat...
the list might go on and go on. I just find a lot of uses for grenadiers, they proably a support unit with largest variety of uses and tasks it can complete in game, just needs some improvisation.

I use Grenadiers as Marines. I attach them to my regular armies in 4 company battalions, and I use them as assault troops and flankers. Once the line is engaged they flank around and hit enemy troops with grenades. They also have greater accuracy, better morale and better Melee stats than the average line unit, so I treat them like elite troops. Really, I have 16 units of grenadiers throughout my empire, and I ship them as named four unit battalions to wherever the action is hottest. India, North Africa, South America, the Baltic, - You name it, so go the marines...
What can I say? Grenadiers are my favorite unit in nearly every game about this era of warfare...
I use grenadiers to flank enemy and toss nades which usually will cause them to rout.
I prefer 2 units on the GC. Flank both units around to the side, and have them run around to just behind a line unit engaged in gunfire, and time both grenade tosses to go off at the same time. On normal difficulty its almost an instant route. It will take a few throws on VH, but you're still killing a ridiculous amount of troops.
In multiplayer i use them in a similar way, although you have to be a bit more careful with your positioning/movement. I prefer early period (seems like there's far less "stupid" things people do) but on late I like 3 units or so of grenadiers. At times I think I under use them, as they'll typically out-score my line infantry (and are much cheaper) in kills. That being said, it does become a lot of micromanagement in multiplayer. They can also be useful for reinforcing areas that are under bayonet/calvary charge in multiplayer as they have an excellent morale/defense score.
These guys are the shock troops. I normally keep 4 in my army. I put one near each flank, behind the line, and two in the center as reserves. Grenades are just *devastating*. I don't know how many times these guys have pulled victory out of the jaws of defeat when a flank started wavering.
Early on I didn't really know how to use them, but now they're an army mainstay.
