How do I make a region off limits or unconquerable?

    there are several ways

+ its Arguin (the AI wont find you there under any circumstances...)
+ you surround the border of the province with dozens of forts with full stack armies in each
+ you put so many armies in it that the AI has to beat them all just to reach the full garrisoned city. (200 peasant units are enough, you loose all battles but until the AI reaches your city, the game should be over )

Go to some unlinked island ( Crete ) and make a huge fleet to defend it.

I Find an amazing way to prevent crossings into your territory from sea is to place a Ship at a land to land crossing, this will prevent armies from crossing the river, The AI dosn't build ships in this case to transport.

I usually keep full stacks on bridges and block land bridges with fleets.

I do sometimes block land bridges with a fleet, mostly to stop religious units from getting into my lands. If an army tries to cross I'll generally move the ships then put them back when they have crossed over.

The sure-fire, old-fashioned way is to conquer every other province.

Medieval II Total War\data\world\maps\base theres a file there called map_ground_types.tga what you do is take the eyedropper tool and take the color of the southern part in africa. The gray is impassable terrain. You take the pencil or brush and paint impassable to the place where the settlement is or around it or w/e.

if you do that, make sure you move any characters/armies that spawn there and also note that it will throw an error in your log about the settlement being unaccessable.

    Code:
    01:27:40.250 [script.err] [error] Script Error in data/world/maps/campaign/imperial_campaign/descr_strat.txt, at line 3901, column 12
You have chosen an invalid tile(287, 9) for the settlement of Arabia.
It will mean not being able to reach this settlement and is a bug.
    For now we'll ignore it.
the game will still play fine though.

It will as long as that region is not listed in your winning conditions, and is not assigned to a faction. There are probably some other things that you need to do too.
