Do you think there will ever make a update to have some unique cities, like different buildings, architecture, etc. For example Jerusalem, Rome, Constantinople, London, Milan.

Maybe in Empire, but not for M2TW.
Yeah definitely not in MTW2, it is enough we have several types of architecture - to be honest I am happy with that ;-)
Yeh, in Empires they could have Big Ben in the background for Lord Protector Cromwell to be ensconsed under. Or the Eifel Tower for Napolean to parade under, eh? No body over here in Texas would think anything is amiss, except for that one bow legged professor at Rice U., but nobody cares what he says.
This has been discussed before, such as making the unique battle maps for Constantinople and Venice, but it isn't possible. Hopefully there will be such an option in Empires.

well the Basileia – Total War mod team are doing it and they have so far made Palermo, Constantinople, Baghdad, Paris, Jerusalem and Cordoba
Would be nice. When I first got RTW, my first TW game, first thing I did was look at Athens, and I couldn't find an Acropolis, and I was sad.
Four,
Northern European
Southern European
Islamic
Mesoamerican
Greek should be a separate architecture al together, unique from either Southern or Northern European...

Well if you argue that you have to argue the different style of architecture in every country and region; its just not possible. They generalised it as much as they could while keeping distinctive features for different areas, and you've got to realise the constraints on developers' time -- if they create different cities for every region, then other, more important features would be flawed or altogether missed.
Hopefully in ETW there will be a bit more variety, such as cities with rivers or built onto hills and cliffs, but otherwise there's plenty of features which require more attention and will be more appreciated.
