Could anyone explain to me what Killsheet is ?

In the SPQR Readme, there is an Excel file named SPQR6.4 Kill Sheet. This file is a spreadsheet used to record your kills for every battle. It adds them up and tells you the total casualties each nation has suffered. It features a graph and fields to record your campaign's start and end years, the faction you played, and the difficulty setting.

but are the data filled automatically, I mean, is there a kind of macro stuff do do that or ?

No, you have to fill it in manually. I usually write results on a paper than when I am done with the game I go add everything up.
