But if I had to take a more realistic approach, I'd say Medieval III. Everyone wants it, and I'm pretty sure CA is aware of this.

WW1. Always wanted a WW1 Total war game.
That war has been under explored for years, and it is ripe for Total war style play.

I would like to see a Far East "Hegemon: Total War" set in the Warring States period of Chinese history.
It is in fact a disgrace that CA hasn't considered this. WHFB makes a better sales pitch, and to be quite honest, I don't complain since I'm a big fan. However, I would have preferred a serious game grounded in reality. Ancient China, or rather, the Far East is the perfect setting, not only because it wasn't yet really covered in TW games, but also because of the sophistication of military doctrines, civil ideologies and political conflicts that were characteristic of the period. Reading Chinese history, there were a lot of things which even to my Western eyes, appear remarkably relevant to modernity.
I absolutely would like to see the campaign of EmpireTW, but without so many bugs and a BAI like Napoleon or FOTS has. That game could have been so great.
Are you ready for THE WRATH OF KHAN! Have the map spread from the far east to the eastern Mediterranean and Europe.

I'd prefer a game based on the Mongols to go only as far as they went instead. But a Mongol Total War would honestly be my favorite game if it were made. I'd just rather they first make a new engine, test it on a China setting then put all their efforts into it on the Mongol game.

A World at War: Total War would be fun, 20th century tactics, from World War I to World War II. Though it would need a different name, because there is a COD: World at War.

My favorites:
1. Total War: Reformation (1517-1697) -- Somewhat European-focused, but could include some areas beyond. An age of bloody religious conflict (worse than the crusades), with tactics and strategy undergoing transformation at the remnants of the Medieval way of war are transformed into proto-modern armies. Starts at the end of Medieval II with Luther post the 95 Theses, ends at the beginning of Empire as the Nine Year War ended in real history.
2. Total War: Caliphate (633-1073) -- Centered around the Middle East, starting with the great contention between the upstart Arabs and the weakened Persians and Romans. However, the game could extend east into the ruins of the Gupta Empire, where Huns and various local strongmen vie for power, or into the barbarian kingdoms of the West, where a new civilization struggles to emerge atop the ruins of the Western Roman Empire, in spite of the raids of Vikings, Magyars, and Arabs. Starts as Abu Bakr consolidates control over the Arab Caliphate, ends as the Gregorian Reforms signal the resurgence of Catholic Europe.
3. Total War: Three Kingdoms (200-280) -- Centered in China, the game beginning as the Han dynasty ends, just prior to Cao Cao's victory over Yaun Shao. In addition to the Shu, Wei, and Wu, various barbarians factions can take advantage of the troubles such as Xianbei successor hordes, Nanman tribes, or even perhaps some Yellow Scarves who manage to reorganize beyond "mildly troublesome rebel scum" status that TW rebels usually achieve.
My least favorites:
1. Total War: Victoria -- This period is interesting from the perspective of grand strategy, but the tactics -- the emphasis of a Total War game -- of the period seem rather boring. The only really viable force on the land battlefield are rifle-armed infantry and some artillery. Naval warfare perhaps showed some more dynamism, with the rising role of steam power and eventually ironclad warships, but I don't think naval warfare can really carry a TW game. (Through WW1, with the rise of tracked vehicles, air power, and such, tactics got more interesting again -- although how well a TW style game can represent that is an open question.)
2. Total War: Aegean -- pre-Hellenistic (before Philip of Macedon and such) Greece. Like Victoria, this strikes me as an age of interesting politics, interesting strategy, and slightly dull tactics, but with hoplites in lieu of riflemen. Becomes more interesting maybe around the time of Epaminondas.
3. Total War: Redux -- something we've seen before like Empire 2 or the like. Meh.
Interesting ones I don't know whether I like or not:
1. Total War: Fantasy -- I'd sort of like to see something other than A Song of Ice and Fire or Warhammer as the basis (think outside the box!), but it could be interesting. Or could be terrible. I don't know.
2. Total War: Modern -- A World War II type game or something could be interesting. Certain the age was an interesting one strategically and tactically from my view, but whether air power and such could be handled with anything like a conventional TW engine is uncertain.
3. Total War: Civilization -- melding a 4x strategy game's "ground up" development from stone age to space age (or whenever to whenever), possibly including randomly generated strategic maps, and including the TW tactical battle game, could be amazing, but I don't know how well it would work.
Undoubtedly, Medieval III with the Renaissance included : from the Rise of Islam to Thirty Years' War (622 -1648).
Empire/Napoleon TW it was an unhappy experiment and I think they will not repeat.
It seems that the producers want to put more emphasis on fighting with swords and shields.I do not think the series will go beyond a nineteenth century.
Also, mechanical TW series does not seem compatible with the era and military tactics and technics of the two world wars.
For Mesopotamia TW it is sufficient a mod for Rome 2.

I vote for Victoria if it had a world map like that of Empire! It'd be awesome to get to fight the American Civil War, Zulu Wars, Crimean Wars as well as a bunch of others.
This is my idea: Dark Age Total War.
Now, we've sort of already had this with Barbarian Invasion and Attila, but I would set the game a couple of hundred years later, starting in 632 and ending in 1090. The map would stretch from Scandanavia to the Equator and from the Atlantic to the Pacific. I have a more advanced map, but it's a work in progress, so I'll show you the more basic map with the rough starting locations of the playable factions. There would be many non-playable factions (though mods could add them if people wished), but the 14 I have selected as playable are:
The Franks, The Visigoths, The Avars, The Byzantine (Or Eastern Roman) Empire, Ghana, Axum, The Caliphate of Mecca, The Sassanians, The Gokturks, Tibet, The Harsha Empire, The Chalukya Dynasty, The Tang Dynasty and Chenla

I would love to see a Medieval III: Total War, with a new engine and up to date graphics. It would progress from early period (1100) to late period (14-1500). Love to see chain mail armies of early medieval times, then transitional armor, full plate, and finally renaissance.
For something new, I'd like to see a 1500-1880 period. From late renaissance up to the Franco Prussian War of 1880. So, it would be pike and shot, then musket and cannon, then the intro of machine guns and bolt action rifles. It would cover the 30 years war, US Civil War, scramble for Africa, and creation of German Empire, to name a few things.
Most importantly, regardless of era, is the impact of a new engine on the gameplay. I can just imagine the realism, battlefield carnage, and campaign map/faction/rpg elements. Let's hope they take their time developing it so we can enjoy something truly reinvigorating.

I would love to see a Total War version of Paradox games, like Victoria, or Europa Universalis. The possibility of playing a fleshed out game as Brazilian Guarani Natives, or as Zulu, or as or as Apaches, really calls for me. I love playing (or at least the idea of playing) as this side of history.
in summary, I want the world to be full of factions, all playable and fleshed out.
